HER2 ADC Clinical Trials and Selected TROP2 Comparison
Inspect six selected registry studies involving HER2-directed antibody–drug conjugates and an ADC-sequencing study that also includes a TROP2-directed agent. The 16 October 2026 capture preserves trial identifiers and intervention names. It does not supply a complete ADC market map or automatically classify antibody targets, linkers and payload chemistry.
The six retained records are NCT03529110, NCT03734029, NCT03255070, NCT05494918, NCT06533826 and NCT04351230. They include DESTINY-Breast03, DESTINY-Breast04, ARX788, JSKN003, TRADE DXd and a withdrawn T-DM1/abemaciclib study. TRADE DXd includes trastuzumab deruxtecan and datopotamab deruxtecan; it is not evidence that every listed intervention targets HER2.
An ADC landscape has several different units
The antibody target, payload, linker, drug product, development programme and clinical trial are separate entities. Counting NCT records measures registered studies in the selected set. It does not directly measure the number of molecular designs or independent competitors. The same ADC can appear in several disease settings or in different combinations, while a single randomised trial can include two ADCs.
The reference selection makes this distinction visible through named studies. DESTINY-Breast03 and DESTINY-Breast04 have different eligibility and comparison contexts. Their presence in one table does not make their populations interchangeable. The ARX788 and JSKN003 records provide additional agent contexts without implying that the six-record selection covers every HER2-directed programme.
Read target and population labels precisely
HER2 expression categories can define different clinical populations and may depend on an assay and scoring convention. Treat the registry wording as a pointer to the complete eligibility criteria. A short chart label cannot capture all prior-therapy, disease-stage and biomarker requirements. Check the full record and the relevant results publication before interpreting a study as evidence for a particular patient population.
The TROP2 comparison comes through the retained ADC-sequencing study rather than a claim to a full TROP2 landscape. An intervention list may also include ordinary cytotoxic agents or non-ADC combinations. Preserve those names and their study roles. Do not convert a broad modality label into a precise payload-class claim that the underlying record has not established.
Retain the limitations of a dated registry capture
The selected source records were retrieved on a stated date, and later registry updates can change status, estimates or descriptive fields. A withdrawn study can remain informative about the historical design space without contributing evidence from treated participants. Keep the operational status and enrolment context attached to any interpretation of its place in the set.
To extend this example, define the target and mechanism boundaries first, then document searches and inclusion decisions. Resolve aliases at the intervention level and retain citations for each mechanistic annotation. That produces a reviewable evidence set. The interactive table is a navigation and inspection tool; it does not replace that curation or supply missing efficacy results.
Equations and variables
Share within the selected records
pgroup = ngroup / Nselected
- ngroup
- Selected study records assigned to the stated category
- Nselected
- Number of retained study records; not the number of assets or the complete market
- pgroup
- Descriptive proportion within this selection
Distinct study count
Nselected = |unique NCT identifiers|
- Nselected
- Number of distinct retained registry records
- NCT identifier
- Registry identity of a study, not a unique drug or payload
Scope and limitations
- No automatic payload-class, linker or drug-to-antibody-ratio classification is provided. Any mechanistic assignment requires a cited source specific to the agent.
- HER2-positive, HER2-low and other eligibility descriptions are not interchangeable. Trial populations and prior treatments must be read in their source context.
- The selected records include completed and withdrawn studies. They cannot establish current recruitment availability or comparative treatment effectiveness without checking the current full record and results.

Technical questions
Are all drugs listed in these studies HER2-directed ADCs?
No. Studies may contain chemotherapy, combination partners or agents directed at another target. TRADE DXd includes a TROP2-directed agent as well as a HER2-directed agent. Read intervention identity and study context instead of assigning one target to every name in a trial.
Can the workspace compare ADC payload classes?
It preserves source intervention names and the native descriptive record fields. It does not infer a curated payload or linker taxonomy. A scientifically defensible payload comparison requires agent-specific source evidence and explicit handling of aliases and investigational compounds.
Can these trials be ranked by apparent response rate?
The initial capture does not create a cross-trial efficacy ranking. Populations, endpoints, follow-up and comparators differ, and results may be unavailable in a registry record. Comparisons need the actual reported outcomes and an appropriate analysis of study design and uncertainty.
