Cell seeding calculator: viable cells per vessel

Suspension and medium per vessel from a measured density and viability, with the counting dilution applied once, area or volume targets and a 1:n split.

Cell seeding and splitting

Use measured inputs and your protocol’s targets. Save the exact setup or export a free preparation sheet.

Select targets for your cell line and experiment. Confluence is not a cell count. Available suspension is required to check the finite allocation.

  • Calculated setup only; this is not a performed preparation or measurement.
  • Targets are supplied by the researcher. Viability and counting dilution are each applied once according to the selected measurement basis. No confluence-to-count conversion or cell-line recommendation is made.
  • Volumes are treated as additive. Overages, when selected in measured mode, scale the complete seeding mixture once; the stated per-vessel target is unchanged.

Illustrative arithmetic only; no biological target or equipment setting is recommended.

Assumptions and qualifications

Open in the bench sheetThe bench sheet carries these inputs and offers the other calculators.

Worked example

  • 2 million cells/mL at 80 percent viability is 1.6 million viable cells/mL; 100,000 viable cells in each of 10 vessels needs 0.625 mL of suspension before overage.
  • With 10 percent overage of the complete mixture: 0.6875 mL. Entering the already-viable density, 1.6 million cells/mL, gives the same answer without applying viability twice.
  • Choose targets for your cell line and experiment; none is suggested here. Confluence is not a cell count.
  • The 1:n split mode divides a harvested suspension into equal parts and makes no prediction of density or growth.

Method

  • Cell seeding from a measured density, and 1:n splitting. Viable density from total density and viability, counting dilution applied once; stock per vessel = target / viable density (Thermo Fisher Gibco cell culture basics, maintaining cultured cells; read 10 October 2026).Checked against exact rational arithmetic (Python 3.13.16 fractions) on 13 cases, five of them refusals, to nine significant figures; refusals exact (10 October 2026).

Limits

  • For research and education only, and not medical advice: the stock, dilution and buffer calculations here are not for dosing or clinical preparation.
  • Cell seeding uses the density, viability and targets you supply; it recommends no density, predicts no growth or confluence, and a 1:n split predicts nothing.
