Antibody dilution calculator
Stock and diluent for a working antibody dilution by concentration or a stated 1:n ratio, for a batch of vessels with your overage and dead volume.
Antibody dilution
Use measured inputs and your protocol’s targets. Save the exact setup or export a free preparation sheet.
Antibody and source details
Answer
Batch: 0.02 mL stock and 9.98 mL diluent, a 1:500 dilution.
- Stock for the whole batch
- 0.02mL
- Diluent for the whole batch
- 9.98mL
Planned calculation; no performed preparation is recorded.
Calculated setup
| Quantity or basis | Value |
|---|---|
| Planned vessels | 1 |
| Final volume per vessel | 10 mL |
| Final volume to make up to | 10 mL |
| Dilution factor | 500 total parts per stock part |
| Final antibody concentration | 0.002 g/L |
| Stock for the whole batch | 0.02 mL |
| Diluent for the whole batch | 9.98 mL |
- Calculated setup only; this is not a performed preparation or measurement.
- C1V1 = C2V2. A 1:n ratio means one stock part in n total parts. Batch volumes sum the entered final volume per vessel, with only the overage and dead volume you choose; a homogeneous mixture and additive volumes are assumed. The researcher or datasheet supplies the target; specificity and optimum staining are not predicted.
Illustrative arithmetic only; no biological target or equipment setting is recommended.
Assumptions and qualifications
Open in the bench sheetThe bench sheet carries these inputs and offers the other calculators.
Worked example
- 1 mg/mL to 2 µg/mL in 10 mL: a 1:500 dilution, so 20 µL of stock made up to 10 mL. A stated 1:500 from the same stock gives the same answer: 1:n means one part stock in n total parts.
- 0.5 mg/mL to 1 µg/mL, 0.2 mL in each of 24 wells with 10 percent overage: 5.28 mL of working dilution, 10.56 µL of stock, by the same overage and dead-volume rule as the master mix.
- The dilution comes from you or your datasheet; nothing here recommends one or predicts specificity. Clone, catalogue and lot are free-text notes.
- Below your minimum transfer, the constrained intermediate planner takes over; ratio-only mode, with no stock concentration, cannot use it.
Method
- Antibody working dilution by C1V1 or a stated 1:n ratio, for a batch with chosen allowances. C1V1 = C2V2; 1:n is one part stock in n total parts (Abcam antibody dilution calculator, read 10 October 2026); batch = N × V × (1 + overage) + dead volume, the master-mix allowance rule. The intermediate path uses the constrained one-intermediate planner.Checked against exact rational arithmetic (Python 3.13.16 fractions) on 20 cases (10 without and 10 with batch allowances), six of them refusals, to nine significant figures; the intermediate path against the brute-force grid of the intermediate-dilution set (10 October 2026).
- Dilution by C1V1 = C2V2. src/lib/dilution/c1v1.ts 1.0.0Checked against Python's exact rational arithmetic (fractions.Fraction) to nine significant figures on 31 fixtures (6 October 2026).
- Constrained one-intermediate dilution. src/lib/dilution/intermediate.ts 1.0.0Checked against an exact brute-force grid search (Python 3.11.15) on 13 cases; the plan within one grid step (10 October 2026).
- Master mix preparation. Volume balance and C1V1 = C2V2; protocol-specific overage and user-supplied single-vessel dead volume. Thermo Fisher MAN0025706 B pp. 12–13; INTEGRA dead-volume technical note, read 9 October 2026.Checked against exact rational volume balance (Python 3.11.15) on 14 cases to nine significant figures (10 October 2026).
Limits
- For research and education only, and not medical advice: the stock, dilution and buffer calculations here are not for dosing or clinical preparation.
- Antibody dilutions use the stock and target, or the 1:n ratio, that you or your datasheet supply; no dilution is recommended and specificity is not predicted.
